TheculturedcorticalneuronswereincubatedwithPio0.01,0.1,1and10μmol·L-1for1h,thenaddingAβ25-3520μmol·L-1for24h.GW9662wasadded30minbeforePio1μmol·L-1added.ThecellviabilitywasassessedbyMTTassay.TheneuronalapoptosiswasquantifiedbyscoringcellspercentagewithapoptoticnuclearmorphologyafterHoechst33258staining.Theproteinexpressionsofphosphorylatedmitogenactivatedproteinkinasekinase4(MKK4),phosphorylatedcJunNterminalkinase1(PJNK1),andBcl2weremeasuredbyWesternblotting.RESULTS ThedecreasingofcellviabilityandincreaseingofapoptoticcellsinculturedcorticalneuronswereobservedinAβ25-35groupcomparedwithnormalcontrolgroup.Thecellviabilityofcorticalneuronswasdecreasedto(66.8±1.2)%inAβ25-35modelgroup(P<0.01).Pio0.1,1and10μmol·L-1canantagonistagainstAβ25-35induceddecreasingofcellviabilityinconcentrationdependentmanner,andwere(81.2±2.9)%,(87.9±2.2)%and(98.1±1.9)%,respectively.GW9662didpartlyinhibiteffectsofPiooncellviability.Aβ25-35inducedincreasingofapoptoticcellpercentagesfrom(11.8±1.1)%innormalcontrolgroupto(64.6±2.3)%(P<0.01).Pio0.1,1and10μmol·L-1preventedincreaseofapoptoticcellspercentage,respectively(58.3±1.2)%,(52.6±1.3)%and(41.5±1.5)%.Furthermore,neuronsincubationwithAβ25-35for24hinducedthedecreaseofBcl2expression,increasesinphosphoMKK4andpJNK1expressions.PretreatmentwithPiosignificantlyinhibitedreductionofBcl2expressioninducedbyAβ25-35.Pio0.1,1and10μmol·L-1alsoinhibitedAβ25-35inducedincreasinginMKK4andJNK1phosphorylation.CONCLUSION
